How to store cells in a liquid nitrogen tank?

Generally speaking, the procedure of cryopreservation of cells will affect the vitality of cells during recovery, so when using liquid nitrogen tank to store cells, you should be careful and follow the steps:
1. Pre prepared cryopreservation solution: 10% DMSO + 90% fetal bovine serum, stored in refrigerator at 4 ℃ for precooling;
2. Digestion of cells with trypsin: pour out the medium in the culture bottle or carefully suck the medium in the culture plate with a gun, wash twice with PBS, digest the cells with trypsin (as mild as possible);
3. Suspend the cells with the complete culture solution again, add the precooled cryopreservation solution to the cells with complete digestion, and gently blow and mix with the burette.
4. Add 1ml of cell solution into each cryopreservation tube and mark the name and date of cryopreservation after sealing.
5. The detailed cryopreservation process is directly related to the vitality of cells during recovery. If there is a program cooler (put it in the refrigerator at - 80 ℃ overnight, put it in the liquid nitrogen tank), it is best; or it can be at 4 ℃ for 2 hours;
To sum up, cells are usually stored at - 80 ℃ for half a year, and can be stored in liquid nitrogen for a longer time.
