1. The slower the freezing, the better the recovery.
The slower the cell freezes, the better, the more time it needs before freezing; the faster the recovery is, the better. After removal from the liquid nitrogen tank, quickly put it in a 37 C water bath, and then shake it vigorously to melt it as soon as possible.
2. Notes for Cell Cryopreservation and Resuscitation
Protective glasses and gloves should be worn in contact with liquid nitrogen. Because of the shaking during the recovery process, the vial may explode; moreover, DMSO in the cryopreservation solution is toxic and carcinogenic. So always wear gloves when contacting.
3. Enough number of frozen cells
No matter how careful you are, cells will die in the process of freezing and resuscitation. So the number of frozen cells should be sufficient at first.
4. Marking
Put the culture bottle in the handle. A small card should be attached at the end, indicating the owner's name, cell name and freezing date. The cell name, algebra and date of freezing should also be written on each tube. Laboratory liquid nitrogen tanks are common. Everyone should have their own handles, which will save a lot of time for finding their own liquid nitrogen tank.
5. Careful Care
The recovered cells are very fragile and need our careful care. All contact with cells should be gentle.
6. Problems of DMSO
As mentioned earlier, DMSO is a toxic carcinogen. So, after thawing, should DMSO be removed from the freezing solution?
It does not need to be removed. Except for a few specifically designated DMSO-sensitive cells, 99% of the cells do not need to remove DMSO. Frequent liquid exchange after thawing will slowly remove DMSO.
7. The substances used in cells before and after thawing should be the same.
Because each cell has its own specific and familiar growth environment. Suddenly using different cultured blood and serum can cause poor cell growth and even death.
Cell culture is a laborious task, which requires constant attention. Always pay attention to whether the cells in the incubator should be changed. This problem can be solved by mastering effective freezing and resuscitation technology.
